چکیده

THE INITIAL barrier to successful organ xenotrans1. plantation in discordant species is hyperacute rejection (HAR) mediated by either natural (preformed) or induced antibodies. 1•2 Hamster cardiac xenografts are normally rejected by this mechanism within 3 days when transplanted into naive LEW or athymic (nude) rats,3 which in the latter is supposedly caused by a T-cell independent process. This is contrary to the prevailing tenet that dictates that T-cell help is required for the initiation of HAR following presensitization with xenoantigens. Accordingly, we hypothesized that prior exposure of T -cell-deficient nude rats to donor antigens would not precipitate HAR of a subsequently transplanted hamster heart. This study was therefore designed to test this hypothesis.

Donor-specific transfusion in the nude rat prolongs survival of subsequently transplanted hamster cardiac xenografts.

XENOGRAFfTUMORS have indefinite survival when transplanted into atbymic nude rats or mice. However, nude rats reject vascularized hamster cardiac xenografts in tempo (3 to 4 days) similar to that of immunocompetent rats.13 This "slow-motion" hyperacute rejection witnessed in the nude rat is thought to be mediated by a T cell-independent, but antibodyand complement-dependent mechanism(s ).1 In t...

Donor species complement after liver xenotransplantation. The mechanism of protection from hyperacute rejection.

Hamster hearts transplanted into stable rat recipients of hamster livers (OLT rats) were hyperacutely rejected after transfer with unaltered rat antihamster hyperimmune serum (HS). This was followed by immediate liver xenograft rejection in 4 of 5 rats. In contrast, simple heat inactivation of the rat HS resulted in prolonged survival of hamster hearts to 25 days without deterioration effect in...

Use of MHC class I or II "knock out" mice to delineate the role of these molecules in acceptance/rejection of xenografts.

I T has previously been reported that pretreatment of human islets with anti-MHC class I antibodies leads to their prolonged survival when subsequently transplanted into naive mice, suggesting that, as in allotransplantation, these molecules also playa role in the recognition and the initiation of effector responses to xenoantigens,l However, this protracted survival of cellular, unlike primary...

Successful hamster-to-rat liver xenotransplantation under FK506 immunosuppression induces unresponsiveness to hamster heart and skin.

In addition to its well-known resistance to antibody (1) and cell-mediated (2) rejection, the liver allograft can confer these advantages to other tissues and organs transplanted concomitantly or subsequently from the donor or donor strain (3, 4). We report here an example of this same hepatic tolerogenicity after hamster livers were transplanted to rats under FK506, ameliorating thereafter the...

Effect of anticomplement agent K76 COOH on hamster-to-rat and guinea pig-to-rat heart xenotransplantation.

In normal rats, the xenobiotic K76 inhibited the C5 and probably the C2 and C3 steps of complement and effectively depressed classical complement pathway activity, alternative complement pathway activity, and the C3 complement component during and well beyond the drug's 3-hr half-life. It was tested alone and with intramuscular tacrolimus (TAC) and/or intragastric cyclophosphamide (CP) in rat r...
